Purification and properties of D-glycerate 3-kinase from Escherichia coli.

چکیده
The adaptive D-glycerate 3-kinase from glycerate-adapted Escherichia coli has been purified 4200-fold in 37% yield and has been crystallized. The procedure, starting with the crude, cell-free extract, consisted of treatment with protamine sulfate, ammonium sulfate, passage through a diethylaminoethyl Sephadex column, and precipitation with ethanol. The reaction product, 3-phosphoglycerate, has been identified by paper chromatography and specific enzymatic assay. Some characteristics of the enzyme which show clearly its differences from other glycerate kinases are described.

D-GLYCERATE 3-KINASE, the last unknown enzyme in the photorespiratory cycle in Arabidopsis, belongs to a novel kinase family.
D-GLYCERATE 3-KINASE (GLYK; EC 2.7.1.31) catalyzes the concluding reaction of the photorespiratory C2 cycle, an indispensable ancillary metabolic pathway to the photosynthetic C3 cycle that enables land plants to grow in an oxygen-containing atmosphere. Except for GLYK, all other enzymes that contribute to the C2 cycle are known by their primary structures, and the encoding genes have been iden...
متن کاملEnterotoxigenic Escherichia coli infection induces tight junction proteins expression in mice
Enterotoxigenic Escherichia coli (ETEC) causes diarrhea in travelers, young children and piglets, but the precise pathogenesis of ETEC induced diarrhea is not fully known. Recent investigations have shown that tight junction (TJ) proteins and aquaporin 3 (AQP 3) are contributing factors in bacterial diarrhea. In this study, using immunoblotting and immunohistochemistry analyses, we found that E...
متن کاملAn In Vivo Metabolic Approach for Deciphering the Product Specificity of Glycerate Kinase Proves that Both E. coli’s Glycerate Kinases Generate 2-Phosphoglycerate
Apart from addressing humanity's growing demand for fuels, pharmaceuticals, plastics and other value added chemicals, metabolic engineering of microbes can serve as a powerful tool to address questions concerning the characteristics of cellular metabolism. Along these lines, we developed an in vivo metabolic strategy that conclusively identifies the product specificity of glycerate kinase. By d...

Hansen, Robert W. (University of Illinois College of Medicine, Chicago) and James A. Hayashi. Glycolate metabolism in Escherichia coli. J. Bacteriol. 83:679-687. 1962.-This study of glycolate-adapted Escherichia coli indicates that the most probable route for utilization of the substrate includes glyceric acid, 3-phosphoglyceric acid, and the tricarboxylic acid cycle. A glyceric acid dehydrogen...
متن کاملCloning and evaluation of gene expression and purification of gene encoding recombinant protein containing binding subunit of coli surface antigens CS1 and CS2 from Enterotoxigenic Escherichia coli
Background & Objective: Enterotoxigenic Escherichia coli (ETEC) is a major causative agent of diarrhea. Enterotoxins and the colonization factors (CFs) are major virulence factors in ETEC infections. The bacterium binds to the intestinal epithelial cell surface through colonization factors and produces enterotoxins that cause excessive fluid and electrolyte secretion in the lumen of the intesti...
